Job Summary

The focus of the Rodrigues Lab is to investigate the switch in energy supply and the mechanisms of cardiac metabolism during diabetes. Applicants must therefore have extensive knowledge and experience in various lab techniques to investigate metabolism including, but not limited to, cell culture (cell lines and primary cell isolations), molecular biology (isolation of DNA and RNA, RT-PCR) and biochemistry (Western blots and enzymatic assays). The Research Assistant/Tech 2 must also have animal handling experience and have previously worked with rodent models. The individual will be expected to perform experiments and data analysis in coordination with other lab members.

We have a fully operational lab with all of the equipment necessary to complete our projects. Our research routinely involves work with hazardous materials, including radioactive chemicals, organic solvents, acids and compressed gases. The laboratory is licensed to use radioactive material and all employees must successfully complete the Biological, Chemical and Radiation Safety courses offered by UBC. All cell culture work will be performed in a biological safety cabinet and animal handling certification is mandatory.

Organizational Status
The incumbent will report to Principal Investigator and the Senior Research Assistant. Must be able to work collaboratively with graduate students in the laboratory.

Work Performed

Following well-established guidelines and procedures, the individual will assist with experiments investigating the effects of diabetes on biochemical and functional changes in the heart.
- Performs routine cell culture work (endothelial cells, primary cardiomyocytes), and prepares samples for downstream assays.
- Prepares reagents and buffers, cell culture media, and performs _in vitro_ experiments.
- Analyses, assesses and interprets protein expression and activity using Western blots, ELISAs, and enzymatic assays.
- Assists with complex procedures on animals including injections, sample collection, and animal monitoring. Comfortably handles rodents and helps to coordinate and conduct long-term experiments.
- Assists in collecting, organizing, and tracking blood and tissue samples from experimental animals.
- Assists in the extraction of DNA and RNA from biological samples and uses molecular biology techniques including RT-PCR to assess RNA expression and genotyping.
- Assist in the planning and conducting of immunofluorescence microscopy and/or immunohistochemistry.
- In collaboration with senior lab members, assists with troubleshooting and the optimization of protocols.
- Assists with presenting, analyzing, and interpreting experimental data including the use of software such as GraphPad Prism. Presents their own findings during weekly lab meetings.
- Performing other general lab related duties as required.

Consequence of Error/Judgement

Failure to report problems could result in lost or unusable data from experiments resulting in delay of project progress, and require that the experiments be repeated.

Errors at any stage may result in loss of precious resources (animals and biological samples) and a misinterpretation of the data. Repeated inaccuracies and errors of the same type are incompatible with the position.

Supervision Received
The incumbent will be supervised by the PI and Senior Research Assistant, but will need to communicate and work closely with graduate students in the lab. Once trained, the Research Assistant/Tech 2 will work mainly independently following established protocols.

High school graduation, some additional training in a related field and a minimum two years of related experience or an equivalent combination of education and experience.
