Job Summary
Organizational Status
The Research Technician reports directly to Dr. Bruce Vallance and Dr Andy Sham and liaises with other members of Dr. Vallance's research group, including co-investigators, researchers, research assistants, post-doctoral fellows, students and staff.
Work Performed
**Providing support with research activities including**:
Process intestinal biopsies for organoid isolation derived from patient samples and maintain collection records
Undertaking cell culture experiments using primary epithelial cells obtained from patient samples
Manage the daily operation of the organoid maintenance including culturing and passaging of intestinal organoid cultures.
Prepare organoid media and performing routine testing to identify possible contamination following SOP
Perform functional analysis of intestinal epithelial cells and undertake experiments related to microbiology and immunology (ELISA, FACs, immunostaining etc)
Communicating with researchers and students the results of cell culture experiments and support in organoid maintenance
Maintain inventory of frozen organoids and perform routine testing for storage conditions Improve and optimize freezing procedure as required for the organoid team
Design and perform primary cell culture experiments for drug testing and bacterial infection and interpret results
Troubleshooting the result and procedures of experiments and revise experimental plan
Develop novel protocols and optimize experimental procedure gut-on a chip experiment on the Emulate organ on a chip system and cell culture under anaerobic condition
Develop operation protocol and SOP for microbiome and cell culture specific equipment ( Emulate organ on a chip system, anaerobic chamber, infection under hypoxic condition)
Maintain a detailed lab notebook
Provide training to undergraduate students, as well as new and lower level technicians in the use of equipment, techniques and procedures such as the organoid on a chip system, and training for developing biopsies samples from patients derived organoids
Set up, test, operate and maintain common laboratory equipment and laboratory reagents for the lab
Consequence of Error/Judgement
The successful applicant will exercise a considerable amount of judgment, responsibility, and initiative in determining work procedures and methods, and coordinating the work of the unit.
Dr. Vallance's laboratory is situated within the Childhood and Disease Theme of the BC Children's Hospital Research Institute. Errors made could compromise the results of research projects, and therefore impact the credibility of the Unit, Centre, Institute and individual Investigator(s).
Supervision Received
The successful applicant will work under mínimal supervision, receiving specific instructions only on unusual problems or on matters which depart significantly from established policy and procedure.
Supervision Given
May oversee and direct the work of one or two employees at lower classifications in which case the successful applicant is responsible for the accuracy, production, and control of the work unit. The successful applicant may also act a resource for junior trainees
Minimum Qualifications
Completion of a university degree in a relevant discipline or technical program and a minimum four years of related experience or an equivalent combination of education and experience. Some positions may require a graduate degree.
