Job Summary

The UBC Bioimaging facility is a multi-user imaging facility that provides access, training and service to users across and beyond UBC. The facility supported more than 226 researchers from 118 research groups including 26 UBC departments and 7 faculties, local hospitals, other universities and industry clients in Fiscal 2024. For this position, the technician requires specialized training and expertise in Transmission Electron Microscopy (TEM), Electron Tomography, High Resolution TEM, Cryo-TEM and TEM sample preparation techniques for both material and biological samples.

The technician will provide technical assistance to graduate students, staff and faculty on TEMs, TEM sample processing equipment and advanced image processing software.

Trains and supervises graduate students, post-doctoral researchers, and faculty how to use the TEM and sample preparation equipment.

Performs sample preparation for TEM and conduct various types of TEM imaging (conventional TEM, Cryo-TEM and Electron Tomography) on the samples submitted by clients from UBC, other academic institutions and industry.

Responsible for the operation and maintenance of three Transmission Electron Microscopes (TEMs) such as FEI Tecnai G2 and FEI Tecnai Spirit and TEM sample preparation equipment such as Leica High Pressure Freezer, Leica Automated Freeze Substitution machine, FEI Vitrobot, Leica microtomes (UCT and UC7) and Leica IGL automatic immunogold labeling system.

Responsible for radiation safety and working with radiation safety officers.

Assists users in acquiring, processing and analyzing electron tomography data using advanced image processing software, such as ThermoFisher Inspect 3D and AMIRA.

Maintains equipment and laboratory supplies in the Bioimaging Facility.

Organizational Status
Reports to the Research Manager, and Botany Administrator for administrative matters. Consults with the Research Manager and the Director and provides technical advice on equipment maintenance and replacement.

Work Performed

Provides technical expertise and consultation on diverse research projects using TEMs.

Responsible for the operation and maintenance of the TEM and sample preparation equipment.

1. Training and supervision

Responsible for training users on the operation of two TEMs (FEI Tecnai G2 and FEI Tecnai Spirit) and the sample preparation equipment (Leica High Pressure Freezer, Leica Automated Freeze Substitution machine, FEI Vitrobot, two Leica microtomes (UCT and UC7) and Leica IGL automatic immunogold labeling system).

Responsible for training users on the advanced imaging analysis software, AMIRA, Inspect 3D for electron tomography.

Provides technical instruction on the settings and methods of the equipment.

Assists undergraduate and graduate students’ TEM projects for their thesis work.

Facilitates student outreach through lab tours and instrument demonstrations.

2. Operation and maintenance of the equipment

Operation and maintenance of two TEMs and the sample preparation equipment.

Performs sample preparation for TEM and TEM imaging (conventional TEM, High Resolution TEM, Cryo-TEM and Electron Tomography) on the samples submitted by the clients from UBC, other academic institutions and industry.

Performs periodic maintenance of vacuum systems, replacement of filaments, troubleshooting, maintenance and cleaning of the equipment including electron gun parts.

Troubleshooting, maintenance and cleaning of the equipment.

Documents all problems and downtime of the TEMs and sample equipment, including symptoms and solutions/modifications made during the repair process. Allow previously encountered problems to be identified more easily.

Responsible for radiation safety and working with radiation safety officer.

Maintains a stock of consumable parts and repair items for TEMs. Informs the Research manager when the purchase of items is required. Assists in the ordering of supplies and equipment.

Works with other Research Technicians on shared duties such as maintaining solvent, chemical and lab consumable stocks, as well as safety and cleanliness of the fume hoods and sinks.

Performs other duties related to the qualifications and requirements of the job.
